Scientists announce decoy-proof Ebola antibodies

Findings published Thursday in Cell reveal the development of a new antibody treatment for the deadly disease caused by the Ebola virus and its relatives.

Scientists from the United States-based La Jolla Institute for Immunology (LJI) and National Institute for Allergy and Infectious Diseases identified two antibodies, 1C3 and 1C11, that can attack Zaire ebolavirus and Sudan ebolavirus.

There are currently no vaccines or antibody cocktails available to treat ebolaviruses other than Zaire ebolavirus. LJI President and CEO Erica Ollmann Saphire said: “Finding antibodies with this breadth is important because we don’t know which virus in the genus of ebolaviruses is going to break out next.”

Of the six ebolaviruses known to science, the Zaire and Sudan ebolaviruses are the two most deadly: the 2010s outbreak that killed eleven thousand was caused by the Zaire virus.

The new contribution made in this paper is that scientists used the imaging technique cryogenic electron microscopy to observe how and where the antibodies interact with the viruses.

The team saw that 1C3, instead of binding to the target site on the viral glycoprotein in the typical molecular lock-and-key fashion, stuck itself sideways across the glycoprotein, blocking three binding sites at once. Antibodies work by binding to pathogens and neutralizing their antigens.

1C11 works by interfering with the molecules that the virus would use to enter a cell. Because the Zaire and Sudan ebolaviruses use very similar mechanisms, 1C11 is effective against both.

According to the research team, the primary advantage that these two antibodies have over others is that they are resistant to cross-reactivity. When some viruses enter the body, including the Zaire and Sudan ebolaviruses, they produce excess glycoprotein and release it into the area like a smokescreen. This causes many of the body’s antibodies bind to this glycoprotein instead, as many as 90 percent of them. 1C3 and 1C11 do not; they bind only to the real virus.

This means that antibody treatments based on 1C3 and 1C11 are likely to be effective at much lower doses than other treatments, as early experiments have already shown: One treatment has already been tested in non-human primates, and even the lowest doses used were 100 percent effective. This also means the human treatment might be much cheaper to produce than expected.

According to LJI, treatment based on these two antibodies would offer two advantages over existing treatments. First, patients with undiagnosed species of Ebolavirus can be treated with an antibody cocktail that is effective against more than one virus. Because doctors don’t always have time to figure out which virus a patient has, this could save many lives. Second, these antibodies would be effective in patients with later stages of the disease.

The experiments were conducted using samples donated to Emory University by two Ebola survivors.

U.S. outlaws online gambling

Over the weekend, millions of Americans faced the new ban of online gambling.

The legislation was attached by Rep. Bill Frist to the Port Security bill.

Many of the public companies that offer online gambling have been quick to react and give their outlooks on the legislature. Party Gaming and 888 among others have said that they will shut down all U.S. operations if President Bush signs the bill into law. This sent online gambling stocks tumbling with Party Gaming leading the way with a 55% decrease in market value.

The people involved are awaiting to see what happens over the next few weeks.

China’s ‘Bandit King’ given life term in ‘massive’ bribery case

Lai Changxing, dubbed the Bandit King, has been given a life sentence for years of smuggling and corruption that added up to billions of US dollars or Pounds sterling. The Chinese court described the values as “massive”.

Lai smuggled goods worth more than £2 billion into Xiamen, bypassing more than £1 billion in import duty. He paid 64 local officials a total of almost £4 million in bribes, giving him effective control of the city from 1995 to 1999. He fled China after becoming a wanted man in 1999 and went to Canada via Hong Kong; the following year, Premier Zhu Rongji said “If Lai was executed three times over, it would not be too much”.

As head of the Yuanhua Group, Lai smuggled in cars, chemicals, oil, cigarettes, and other goods. He distributed bundles of cash to the poor, owned and played for his local football team, built stadia, owned a bulletproof Mercedes that once belonged to President Jiang Zemin, and attempted to construct a tower that would have been the nation’s tallest building. He attained local popularity for funding construction projects including schools, hundreds of tower blocks, and the local airport.

As well as money, officials were offered alcohol and prostitutes. Many were offered time at Lai’s seven-storey brothel, the Red Mansion, and feasted at a replica of the Forbidden City.

State TV has broadcast footage depicting a banquet table with a tiger skin laid upon it, cars given to officials, a young woman alleged to have been donated as a lover, and a sackful of gold rings. The case’s prominence was such that Liu Liying, boss of the national Central Discipline Inspection Committee, took charge of bringing Lai down.

Subsequent investigations have examined more than 1,000 suspects with police at one stage turning over an entire hotel to the probe, filling rooms with suspects. National newspaper The People’s Daily has suggested it is the most serious economic crime in modern Chinese history. He was the nation’s top car importer and one of the main traders in oil and imported cigarettes.

Hundreds of officials have been convicted and it is estimated hundreds more remain. Fellow life-sentence prisoners from the case include the city’s deputy mayor and its head of customs. The nation’s former vice-minister for security, Li Jizhou, has a suspended death sentence. Other suspects have killed themselves.

The sums involved are unusually large, and the details are extraordinarily serious

Upon his escape from the nation Lai became China’s most-wanted fugitive. Twelve years of negotiations ended with a Chinese promise Lai would be spared the death penalty, and Canada extradited him last year. Numerous lower-ranking members of Lai’s empire have already been given life imprisonment or death sentences. With execution off the table, the court gave Lai the highest sentence possible: in addition to the life term, he received fifteen years for bribery and had all his possessions confiscated.

The court justified the “double sentence” on the grounds “the sums involved are unusually large, and the details are extraordinarily serious”. “The crimes involve massive sums and particularly serious circumstances,” court officials told Xinhua. Lai had denied corruption at his trial, although he accepted exploiting loopholes to avoid import duty.

“I don’t have a good family background,” Lai said previously in a press interview. “I have to do things step by step by myself. That’s how people came to respect me. I never fussed about big money.” Lai was born as one of eight siblings in the midst of famine.

Demonstrators protest Condoleezza Rice’s trip to Australia

Anti-war demonstrators in Sydney, Australia on Thursday dubbed U.S. Secretary of State Dr Condoleezza Rice a “war criminal” and “murderer.” Two protesters were evicted and five people were arrested during protests against the U.S.-led invasion of Iraq.

Dr Rice, on a three-day trip to Australia, said she understood why people found it hard to be positive about Iraq when all they saw on their television screens was violence.

Soon after Rice began her speech at the University of Sydney’s Conservatorium of Music, two protesters shouted from the rear of the auditorium, “Condoleezza Rice, you are a war criminal,” and “Iraqi blood is on your hands and you cannot wash that blood away.” Standing with their palms towards her, the young man and woman repeated their accusation until security intervened to remove them from the hall.

About 15 minutes into Rice’s address, a third protester appeared at a balcony door, interrupting her speech as she referred to freedom. “What kind of freedom are you talking about? You are a murderer,” said the demonstrator before he was quietly escorted from the hall. “I’m very glad to see that democracy is well and alive here at the university,” she said.

In her speech, Rice sought to justify the U.S. occupation of Iraq, describing Iraqis as now more free. One student asked about abuses committed by U.S. forces at the Abu Ghraib prison in Iraq. She said the abuses had made her “sick to her stomach.” However, she defended Guantanamo Bay, Cuba, where human rights groups say detainees are held in inhumane conditions and in detention flouting international laws.

Before Rice began her speech, about 50 protesters were gathered at the front gates of the Conservatorium. The group were confronted by police on horseback and by police dogs. Police used the horses to charge into the group of activists and push them back, as a police helicopter hovered.

A police spokeswoman said the group was blocking pedestrian access to the building and that police had spent more than 20 minutes warning them to move. The police then moved in and pushed the crowd back 20 metres. Police say five people have been charged with “hindering police in the execution of their duties.”

The “Stop the War Coalition” says Rice is a “war criminal” and is not welcome in Australia. The group’s spokeswoman, Anna Samson, says the protest is one of many planned in the lead-up to the third anniversary of the invasion and occupation of Iraq on March 20.

Paddy Gibson, from the University of Sydney’s Student’s Council, says the protest is in opposition to the Iraq war, and to the use of the University of Sydney’s campus to host Rice, “the most powerful woman in the world,” who they say is a war criminal. “They’re saying, ‘… you’ve got Sydney Uni’s support to stand up and peddle your murderous hate speeches,’ which is what we see it,” he said.

“You’ve got 180,000 people killed, as we said, for no other reason than strategic control of the region’s oil resources. And the anti-Muslim racism that’s been whipped up to justify this war is being felt by Sydney University students,” said Mr Gibson.

North Korea fires balistic missile from submarine

North Korea test fired a ballistic missile from a submarine yesterday, which landed in the Sea of Japan after traveling approximately 500 km (about 300 miles), according to officials of South Korea and the US.

The missile was fired from a submarine off North Korea’s east coast near Sinpo, officials said, and was reportedly North Korea’s first successful launch after missiles only traveled a small distance in previous tests. The South Korean military accused the North Korean government of using the test to increase military tension during the annual South Korean–US joint military drills, which involve 80,000 South Korean and US troops. North Korea has threatened a preemptive nuclear strike saying the drills were practice for an invasion.

This came on the same day as a meeting between the leaders of China, Japan, and South Korea where, according to Foreign Minister Fumio Kishida of Japan, they “urge[d] North Korea to exercise self-restraint regarding its provocative action, and to observe the UN Security Council’s resolutions”. Japanese Prime Minister Shinzo Abe called the test’s intrusion into Japan’s air defense identification zone “a grave threat to our country’s security.”

Scientists report chemotherapy cocktail may cause adult women to grow new egg cells

Chemotherapy is usually associated with a collection of side effects ranging from digestive problems to hair loss, but a study published this week in Human Reproduction demonstrated that female cancer patients may find they have something in common with much younger women in one specific area — their ovaries.

Scientists from the University of Edinburgh examined donated ovarian tissue from fourteen female cancer patients, most of whom had Hodgkin lymphoma, and compared it to tissue from healthy women. They found the samples from women who had been treated with a specific chemotherapeutic regimen known as ABVD not only contained greater numbers of dormant ova — egg cells — than those from women treated with harsher regimens but also more than samples from healthy women. ABVD is named for combining several drugs known as adriamycin, bleomycin, vinblastine, and dacarbazine.

These reproductive cells were not merely more plentiful in ABVD patients. They also appeared immature, “new” in the words of lead researcher Evelyn Telfer. This challenges the conventional belief that girls are born with all the ova they will ever have and the numbers can only go down as the cells are either used up by the reproductive cycle or succumb to damage or natural aging. However, further research is needed to confirm this. The study covered relatively few patients by scientific standards, and David Albertini of the Center for Human Reproduction in New York has suggested the cells may not actually be freshly grown. Instead, they may have always been there and were merely rendered more detectable by ABVD treatment.

The ability to grow new egg cells may have significant implications for women in Western societies, many of whom postpone childbearing to establish careers, sometimes into their late thirties or forties. However, Telfer warns against making use of these findings too soon: “There’s so much we don’t know about the ovary. We have to be very cautious about jumping to clinical applications.”

The experiments had been discussed earlier this year at the annual conference of the European Society of Human Reproduction and Embryology.
